Complement activation profiles in disease.

چکیده

Complement is an acute-reacting non-specific effector system. Activation and consumption of individual components of this multicomponent system occur secondarily to specific complement fixing events. The complement system is therefore an indirect and imprecise diagnostic tool and can be used clinically only as a general indicator of the severity of a particular disease or as a monitor of its progress. An exception is the case of the very rare inherited complement deficiencies where examination of the system does become directly diagnostic. The 11 proteins of the classical pathway, three additional ones of the alternative pathway, and three natural inhibitors can be measured either immunochemically using the appropriate specific antibody or functionally by the appropriate haemolytic assay. At present only a limited number of activation and consumption profiles can be considered to be clinically useful.

Complement factors in Acute Myocardial Infarction and Unstable Angina

Background: Coronary artery disease (CAD) is one of the most important and lethal diseases in the world. CAD represents a board spectrum of disease from silent ischemia at one end to sudden cardiac death at the other end. The middle of this spectrum consists of acute myocardial infarction (AMI) and unstable angina pectoris (UA). Recent data show that the inflammatory process plays a major r...

A Flowcytometry Study of CD55 and CD59 Expression on Erythrocytes in Rheumatoid Arthritis Patients

Background: Inappropriate activation or blockage of the inhibition of complement system could cause tissue damages in autoimmune diseases particularly rheumatoid arthritis (RA). Defect in complement component regulation may cause damages to tissues, on the other hand, or the damaged tissue might affect the unnecessary activation of complement components.   Objective: To investigate the expressi...

Protective immune responses against West Nile virus are primed by distinct complement activation pathways

West Nile virus (WNV) causes a severe infection of the central nervous system in several vertebrate animals including humans. Prior studies have shown that complement plays a critical role in controlling WNV infection in complement (C) 3(-/-) and complement receptor 1/2(-/-) mice. Here, we dissect the contributions of the individual complement activation pathways to the protection from WNV dise...

Detection of complement activation on antigen microarrays generates functional antibody profiles and helps characterization of disease-associated changes of the antibody repertoire.

Humoral immune responses are traditionally characterized by determining the presence and quality of Abs specific for certain Ags. Arraying of large numbers of Ags allows the parallel measurement of Abs, generating patterns called Ab profiles. Functional characterization of these Abs could help draw an even more informative map of an immune response. To generate functional Ab profiles we simulta...
